Ok I installed these cards this morning and this is my update:

Had a PVR-350, 2xPVR-250(15)s, and a PVR-USB2 tuner.
In Sage I removed all the sources then quit Sage and stopped and disabled the service.
Then I ran the hcwclear that came on the CD for the 500 and let it remove everything.
Shutdown. Pulled the 350 and a 250 out and inserted both 500s. Unplugged the usb cable from the USB2 tuner and started up the PC.

Windows detected on bootup and just pointed the hardware wizard to the 500 CD which contains drivers for the pretty much all of Hauppauge's PVR cards except for the 350 which may have been in there too but didn't notice. The drivers installed beautifully then I connected the usb cable back into the USB2 tuner. (It was probably unneccessary for me to unplug it in the first place)
Pointed the hardware wizard once again to search the CD and let it install. It wanted to replace one newer file with an older one for the USB2 but I made it keep the newer file. Then rebooted. Then started up the service etc and Sage and added all the tuners and tested them. Was taping 6 shows at once without a hitch. Just for good measure turned off Sage and rebooted once more. Everything seems to be groovy.
So I'm pretty happy.
On a side note the cards are larger than the 350 or 250s in length. Just something to be aware of when building a system.

When removing all sources, doesn't that remove all of your favorites? My concern is having Sage delete all of my shows if I remove all sources.

Just to add to "Nope"... Favorites are source-independent, are stored in wiz.bin, and are not removed when any source is removed. If a Favorite is limited to a certain channel & you remove that channel, you won't get any new recordings, but the fav is not removed. Alternately, if you add a new source, your favs will be able to be recorded from any new channels (unless, of course, it is limited to a certain network/channel).

Removing sources should have no effect on your existing recordings.

I followed the link provided to shopblt and noticed in the description that the card comes with only the MCE 2005 drivers. Does this mean it will only work on the MCE edition of Windows?
