What is the purpose of this parameter in the HD100 client.properties file?

I understand that when used like the following in the SageClient

media_player_plugin_class/iso=myClass

Sage will attempt to load

myclass

and cast is to the MediaPlayer interface and use that class to provide custom playback for files with that extension.

Which brings me back to my question, in the HD100 client properties those properties have no values (does this mean functionality is built-in) and Sage ignores it?

What are the possible set of properties? I have tried to remove some (with the server stopped) but they are re-added once the server / extender is used.

There haven't been very many mediaplayer plugin classes, and yes, as you supposed, they do default to the built-in sageplayer (or dshow player if so selected in detailed setup) if left blank. As far as I know, the only mediaplayerplugin class that ever got much use is/was the winamp plugin, which I still use for music playback.
